1. state of science: For the investigation of environmental probes exists not a lot of experience and only a few standardized methods. New test methods should be examined for their suitability for the waters control and be developed. The aim of the project no. 6 - comet assay with mussels - was to investigate, optimize and valuate the suitability of the comet assay for testing genotoxic effects in a water relevant organism. 2. method: The water relevant organism, which lives in waters and has an important role for the ecology of waters, Dreissena polymorpha, was used as test-organism. A test method has been developed which allows the investigation of probes and substances for detecting of genotoxic activites in vitro with primary cells of gills. 3. results: In the first phase of the project the testsystem was validated by testing substances and by realization of a ring-test. The testing of probes from the rivers rhine and elbe and some further probes for their genotoxic activity resulted in only one genotoxic probe. Genotoxicity was not obtained by the In vivo-monitoring in a station for online-monitoring of the river rhine near Karlsruhe. 4. conclusion: the comet assay is a suitable method for the controlling of waters. The sensitivity can be improved by further modification of the test method. The comet assay fulfills all criterias for the suitability of biological test methods like relevance, sensitivity, practicability, reproducibility and standardization. 5. applicability: The test with cells of gills of Dreissena polymorpha is useful for examine specific questions. The developed method can be used for a rapid testing of probes for their genotoxicity. The further harmonisation of the comet assay method is necessary for the comparison of results of different working groups.
