Abstract
Chronic opiates exposure is responsible for genes expression alterations. In some brain diseases, there is a correlation between gene expression in CNS and in peripheral blood cells. The aim of our work is to investigate this correlation in morphine treated rats. Microarray study revealed 812 differentially expressed gene transcripts in PBMCs of morphine-dependent rats. The regulation of about 20 selected genes 12h and 36h after the last injection was validated. We then investigated the expression of some of these genes (Adcy5, Gng11, Mir16, Rgs10, Gprk5) in 4 brain regions involved in drug dependence: nucleus accumbens, striatum, prefrontal cortex and hippocampus. The increased expression of Gprk5 in PBMCs was confirmed in the frontal cortex and the nucleus accumbens by in situ hybridization. Morphine treatment induces gene expression patterns in PBMCs similar to reported gene expression in brain and focused on Gprk5 gene expression, which is involved in morphine-dependence mechanisms ;
