Abstract

Abstract A simple procedure for the purification of human J-chain from completely reduced and alkylated polymeric serum IgA or IgM. It was found that the J-chain remained soluble upon dialysis against non-dissociating aqueous solvents, whereas the major part of the heavy and light chains became insoluble and could be removed by centrifugation. Pure J-chain could be obtained from the above supernatant in the second peak from a gel-filtration on Sephadex G-100. This simple procedure has successfully been applied to the isolation of rat J-chain from polymeric myeloma IgA and IgM.
