Abstract

Abstract A method has been developed for the rapid determination of the energy of activation ( E a ) for enzymatic reactions. The basis of the method is that after initiation of the reaction at a lower temperature, the sample is placed in surroundings having an elevated temperature. Temperature and reaction rate are simultaneously determined during the subsequent approach to thermal equilibrium, and E a obtained from the slope of a plot of in rate vs. 1 T . Activation energies determined by this method are reported for the reactions catalyzed by rat brain hexokinase (ATP: d-hexose 6-phosphotransferase, EC 2.7.1.1) and citrate synthase (citrate oxaloacetate lyase (CoA-acetylating), EC 4.1.3.7), rabbit muscle pyruvate kinase (ATP: pyruvate phosphotransferase, EC 2.7.1.40), and phosphodiesterase-I (orthophosphoric diester phosphohydrolase, EC 3.1.4.1) from muskmelon seeds.
