Gamma globulin interactions in the sera of 2 patients with rheumatoid arthritis studied by gel filtration

Abstract

Abstract The technique of gel filtration using polymerized dextran was applied to a study of the interactions of 7S and 19S γ-globulins in the sera of 2 patients with Stage IV peripheral rheumatoid arthritis. A protein mixture consisting of 7S γ-globulin and 17S (uncorrected) rheumatoid factor in a ratio of approximately 6 molecules to 1 was isolated and the constituents were separated by column chromatography. Characterization of the isolated members by serologic, immunoelectrophoretic, and analytic ultracentrifugal techniques, as well as by solubility properties, suggests considerable homogeneity of these interacting γ-globulins. The basis for a specificity in this type of protein-protein interaction is discussed.
