Abstract

Abstract The isolation and identification of androsta-4,16-dien-3-one from human adult male plasma, in both “free” steroid and “sulphate-conjugate” fractions, is described. The identification depends on the method of extraction and purification, including a modified Girard separation, and comparison of the thin-layer and gas-liquid chromatographic properties of the free steroid, its methyl oximes, its enol heptafluorobutyrate and the chloromethyldimethylsilyl ether of the product of reduction with borohydride, with those of authentic androsta-4,16-dien-3-one and its derivatives. The concentration of androsta-4,16-dien-3-one in male plasma is roughly estimated to be 0.01–0.05 μg/100 ml. as “free” steroid and about twice as much in the “sulphate-conjugate” fraction.
