Abstract

Abstract The technique of chemical α-oxidation has been extended so that the specific radioactivities of all carbons, except the five methyl terminal carbons, within a fatty acid such as palmitic acid can be determined. Improvements include the preparation of butyl esters rather than methyl esters of the products of chemical α-oxidation and the use of temperature-programmed radio gas chromatography for the determination of specific activities. A major series of by-products has been identified as dicarboxylic acids, and the mechanism of chemical α-oxidation is discussed. Evidence indicating that [ U- 14C]palmitic acid is indeed uniformly labeled is presented.
