Abstract

Abstract An automated procedure for the precolumn derivatization of amino acids with dansylchloride and a liquid chromatographic method for separation of the derivatives with fluorimetric detection in the picomole range are reported. The method involves a simple solvent preparation, which does not require the inclusion of non-volatile buffers of low ionic strength and delicate pH adjustments. The procedure was also utilized for the identification of COOH-amidated amino acids released from peptides after digestion with carboxypeptidase.
