Electron microscopic study of membranes and periodic structures applying the through-focusing method

Abstract

The through-focusing method was applied to an electron microscopic study of cross sections of evaporated carbon film and nerve myelin by taking pictures at different focal lengths. It was found that underfocus of more than 0.2 μ and any overfocus produces artifacts due to interference of electrons passing through the specimen. These results show that for the study of the molecular structure of biological material, the focal range must be from the theoretical in-focus to 0.2 μ underfocus.
