Abstract

Abstract Various methods of determining protein in cerebrospinal fluid were compared. Of the methods investigated (Kjeldahl, sulphosalicylic acid and Folin-copper methods, and direct measurement in UV light), the spectrophotometric Folin-copper method proved to be the most suitable for routine determinations, because of its high accuracy and the small amount of liquid required. An attempt was made find the reasons why direct measurement in UV light cannot be considered as reliable, and the results were discussed. It was possible to relate the lack of constancy of the extinction of fresh untreated spinal fluid in the region 260–280 mμ, and the disappearance of the peak at 265 mμ after acidification, to the behaviour of ascorbic acid.
