目的: 探究临床常用不同作用机制的保肝药物对X线照射后人肝实质细胞HL-7702增殖的影响,为放射性肝脏损伤治疗过程中保肝药物的选择提供初步的理论依据。 方法: 采用6 MV X射线,10 Gy的照射剂量对人肝实质细胞HL-7702细胞系进行照射,分别在照射前、照射后24、48、72 h于倒置显微镜下观察细胞形态,采用四甲基偶氮唑盐(MTT)法测定细胞存活率,评价射线对肝实质细胞HL-7702的作用。选用保肝药物作用机制中最可能与放射性肝脏损伤发生机制有关的三类及其代表性药物,根据临床用药稀释倍数及相关参考文献,用全培养基稀释成相应的浓度梯度后作用于射线损伤的细胞,MTT法筛选出有效浓度,然后进行药物效果的对比。对射线损伤的肝细胞增殖修复效果明显者检测超氧化酶歧化酶(SOD)、丙二醛(MDA...
