L introduction d acides nucl iques dans des cellules cibles pr sente un int r t pour le d veloppement de nouvelles strat gies th rapeutiques visant diff rents types d affections e g maladies monog niques cancers La famille des lipophosphoramides d velopp e au laboratoire a montr des efficacit s de transfection int ressantes tant in vitro qu in vivo tout en pr sentant des toxicit s mod r es Les objectifs de ces travaux de th se taient de synth tiser et caract riser de nouveaux phospholipides qui tout en conservant une structure similaire aux phosphoramides pr c demment tudi s pr senteraient des variations structurales originales bas es sur les propri t s de fonctions soufr es Afin d am liorer l efficacit de transfection des vecteurs et de mieux comprendre les relations structure-activit qui r gissent les m canismes du transfert de g nes deux approches ont t envisag es Une nouvelle famille de vecteurs les thiophosphoramides a t labor e dans le but de moduler les interactions intermol culaires notamment les liaisons hydrog nes impliqu es dans la stabilisation des complexes ADN-vecteur Apr s des tudes physico-chimiques ces compos s ont t test s en transfection in vitro La deuxi me approche consiste d velopper des vecteurs bio-sensibles afin de favoriser la lib ration de l ADN dans la cellule et de diminuer la toxicit des compos s Une famille de vecteurs comportant des ponts disulfures clivables en milieu r ducteur a t synth tis e et tudi e The use of genetic materials as a drug offers new perspectives for the treatment of certain diseases e g monogenic disease cancer Interestingly some lipophosphoramidate derivatives developed in our group have been identified for their promising in vitro and in vivo transfection activities and their low toxicity The aim of this PhD research was to synthesize and characterize new lipophosphoramidate analogues including sulfured function s in their scaffold In order to improve transfection activities and to have a better understanding of the relationship between structure and transfection efficacies two approaches were explored First thiophosphoramidate derivatives were synthesized to induce a modulation of intermolecular interaction strength in the DNA-lipid complex hydrogen bonds After physico-chemical characterization transfection activities of these compounds were tested in vitro Secondly phosphoramidate analogues with a disulfide bond in their scaffold were synthesized and studied to provide bioresponsive vectors in the reductive intra-cellular environment BREST-BU Droit-Sciences-Sports 290192103 Sudoc Sudoc France FR
